Main Text​

Since the discovery of HIV-1 proteinase in the mid-1980s, this enzyme probably has become the best-characterized peptidase, with several hundreds of crystal structures of inhibitor complexes determined to date. However, the main proteinase of SARS coronavirus (Mpro, also called 3CLpro) has a good chance to catch up. When the first three-dimensional structure of a coronavirus Mpro was published in 2002, that of the enzyme from transmissible gastroenteritis virus (TGEV) [

], interest in the coronaviridae was only moderate, since these viruses were considered relatively harmless to human health. This assessment changed dramatically in late March, 2003, when it was found that the ongoing global outbreak of the highly infectious severe acute respiratory syndrome was caused by a new coronavirus, subsequently named SARS-CoV. Since then, efforts to discover anti-SARS drugs have been numerous, in order to be prepared should the virus raise its ugly head again.
Coronavirus genomes code for two large polyproteins, pp1a and pp1ab, that are processed by viral proteinases to yield the individual components of the large replicase complex. Most coronaviruses have three cysteine proteinases that are responsible for this processing: two papain-like proteinases and the main proteinase, Mpro. Interestingly, one of the two papain-like proteinases is absent in the SARS coronavirus, and the other one has been shown to have the additional function of a deubiquitinating enzyme [

While the papain-like proteinases together are only responsible for three cleavage reactions near the N terminus of the polyproteins, the Mpro cleaves these huge substrates (molecular masses of 450–750 kDa) at no less than 11 sites. Since this reaction is essential for viral replication, the main proteinase is obviously a prime target for interference by inhibitors [

].
As a first step toward inhibitor design for coronavirus Mpros, the crystal structure was elucidated for a complex between the TGEV enzyme and a hexapeptidyl chloromethyl ketone inhibitor that had an amino acid sequence corresponding to the specificity of the enzyme [

]. The mode of binding of this inhibitor to the target enzyme was found to be related (although not identical) to what had been seen earlier in a complex between the rhinovirus (HRV-2) 3C proteinase and compound AG7088 (Figure 1A), a vinylogous ethyl ester developed by Agouron Inc. (now a division of Pfizer) for the treatment of the common cold caused by rhinoviruses [

]. This observation led to the proposal that AG7088 should be a good starting point for the design of anti-SARS inhibitors [

].

Formulating an effective HIV vaccine remains a formidable challenge despite nearly 3 decades of intense research since the virus was first isolated. One of the obstacles that need to be surmounted is the design of a preparation that elicits a potent and broadly neutralizing antibody (nAb) response, i.e. antibodies with the capacity to block infectivity of the genetically diverse pool of HIV strains that circulate globally. The primary target for nAbs on HIV-1 is the envelope glycoprotein spike (Env). Early work on elucidating the exposure of antibody epitopes on Env suggested highly restricted accessibility of antibodies to epitopes that are conserved among otherwise diverse virus isolates. Crystal structures of Env-derived antigens, most in complex with antibodies, along with structure-function studies and molecular modeling, have provided significant further insight into features of Env that limit broad antibody recognition. Despite these challenges, recent progress on various fronts has led to a growing sense that Env is not as impenetrable to nAbs as might have been believed in the past. Increased understanding of antibody epitope exposure on Env should provide new impulses for efforts to elicit nAbs that can protect against HIV-1 infection.

Measles is one of the most infectious diseases with a high mortality rate worldwide. It is caused by the measles virus (MeV) which is a single stranded RNA virus with genetic diversity based on the nucleoprotein gene, including 24 genotypes. In Gabon, several outbreaks occurred in the past few years, especially in 2016 in Libreville and Oyem. A surveillance network of infectious diseases highlighted a measles outbreak which occurred in the south of Gabon from April to June 2017.

Methods​

Clinical specimens of urine, blood, throat and nasal swabs were collected in the two main cities of the Haut-Ogooue province, Franceville and Moanda. Virological investigations based on real-time polymerase chain reaction for molecular diagnosis and conventional PCR for genotype identification were done.

Results​

Specimens were collected from 139 suspected measles patients. A total of 46 (33.1%) children and adults were laboratory-confirmed cases among which 16 (34.8%) were unvaccinated, 16 (34.8%) had received one dose, and 11 (23.9%) had received two doses of the measles vaccine. Phylogenetic analysis revealed that all the sequences of the nucleoprotein gene belonged to genotype B3.

Conclusions​

Measles infection was more commonly confirmed among those with one recorded dose compared to suspect cases with none, unknown or two recorded doses. The molecular characterization of the strains showed the circulation of the B3 genotype which is endemic on the African continent, thirty years after the B2 genotype was described in an outbreak in Libreville, the capital of Gabon. Despite the mesocorticolimbic dopaminergic pathway being one of the main substrates underlying stimulating and reinforcing effects induced by psychostimulant drugs, there is little information regarding its role in their effects at the immune level. We have previously demonstrated that acute exposure to amphetamine (5 mg/kg, i.p.) induced an inhibitory effect on the splenic T-cell proliferative response, along with an increase in the methionine(met)-enkephalin content at limbic and immune levels, 4 days after drug administration. In this study, we investigated if a possible dopamine mechanism underlies these amphetamine-induced effects by administering D1 and D2 dopaminergic antagonists or a dopaminergic terminal neurotoxin before the drug. Pre-treatment with either SCH-23390 (0.1 mg/kg, i.p.) or raclopride (0.1 mg/kg, i.p.), a D1 or D2 dopaminergic receptor antagonist, respectively, abrogated the effects of amphetamine on the lymphoproliferative response and on met-enkephalin levels of the spleen. The amphetamine-induced increase in limbic met-enkephalin content was suppressed by SCH-23390 but not by raclopride pre-treatment. Finally, an intra-accumbens 6-hydroxy-dopamine injection administered 2 weeks previously prevented amphetamine-induced effects on the lymphoproliferative response and on met-enkephalin levels in the prefrontal cortex and spleen. These findings strongly suggest that D1 and D2 dopaminergic receptors are involved in amphetamine-induced effects at immune level as regards the lymphoproliferative response and the changes in spleen met-enkephalin content, whereas limbic met-enkephalin levels were modulated only by the D1 dopaminergic receptors. In addition, this study showed that a mesolimbic component modulated amphetamine-induced effects on the immune response, as previously shown at a behavioral level.

Study of the function of epitopes of Mycobacterium tuberculosis antigens contributed significantly toward better understanding of the immunopathogenesis and to efforts for improving infection and disease control. Characterization of genetically permissively presented immunodominant epitopes has implications for the evolution of the host–parasite relationship, development of immunodiagnostic tests, and subunit prophylactic vaccines. Knowledge of the determinants of cross-sensitization, relevant to other pathogenic or environmental mycobacteria and to host constituents has advanced. Epitope-defined IFNγ assay kits became established for the specific detection of infection with tubercle bacilli both in humans and cattle. The CD4 T-cell epitope repertoire was found to be more narrow in patients with active disease than in latently infected subjects. However, differential diagnosis of active TB could not be made reliably merely on the basis of epitope recognition. The mechanisms by which HLA polymorphism can influence the development of multibacillary tuberculosis (TB) need further analysis of epitopes, recognized by Th2 helper cells for B-cell responses. Future vaccine development would benefit from better definition of protective epitopes and from improved construction and formulation of subunits with enhanced immunogenicity. Epitope-defined serology, due to its operational advantages is suitable for active case finding in selected high disease incidence populations, aiming for an early detection of infectious cases and hence for reducing the transmission of infection. The existing knowledge of HLA class I binding epitopes could be the basis for the construction of T-cell receptor-like ligands for immunotherapeutic application. Continued analysis of the functions of mycobacterial epitopes, recognized by T cells and antibodies, remains a fertile avenue in TB research.

Rotavirus (RV) primarily infects enterocytes and results in severe diarrhea, particularly in children. It is known that the host immune responses determine the outcome of viral infections. Following infections, interferons (IFNs) are produced as the first and the main anti-viral cytokines to combat the virus. Here we showed that RV predominantly induced type III IFNs (IFN-λ1), and to a less extent, type I IFNs (IFN-α and IFN-β) in human intestinal cells. However, it did not produce detectable IFN proteins and thus, was not sufficient to inhibit RV replication. In contrast, we revealed the essential roles of the basal IFN signaling in limiting RV replication by silencing STAT1, STAT2 and IRF9 genes. In addition, exogenous IFN treatment demonstrated that RV replication was able to be inhibited by all types of IFNs, both in human intestinal Caco2 cell line and in primary intestinal organoids. In these models, IFNs significantly upregulated a panel of well-known anti-viral IFN-stimulated genes (ISGs). Importantly, inhibition of the JAK-STAT cascade abrogated ISG induction and the anti-RV effects of IFNs. The aging brain is characterized by altered dopamine signaling. The amino acid tyrosine, a catecholamine precursor, is known to improve cognitive performance in young adults, especially during high environmental demands. Tyrosine administration might also affect catecholamine transmission in the aging brain, thereby improving cognitive functioning. In healthy older adults, impairments have been demonstrated in two forms of response inhibition: reactive inhibition (outright stopping) and proactive inhibition (anticipatory response slowing) under high information load. However, no study has directly compared the effects of a catecholamine precursor on reactive and load-dependent proactive inhibition. In this study we explored the effects of tyrosine on reactive and proactive response inhibition and signal in dopaminergically innervated fronto-striatal regions. Depending on age, tyrosine might lead to beneficial or detrimental neurocognitive effects. We aimed to address these hypotheses in 24 healthy older human adults (aged 61–72 years) using fMRI in a double blind, counterbalanced, placebo-controlled, within-subject design. Across the group, tyrosine did not alter reactive or proactive inhibition behaviorally but did increase fronto-parietal proactive inhibition-related activation. When taking age into account, tyrosine affected proactive inhibition both behaviorally and neurally. Specifically, increasing age was associated with a greater detrimental effect of tyrosine compared with placebo on proactive slowing. Moreover, with increasing age, tyrosine decreased fronto-striatal and parietal proactive signal, which correlated positively with tyrosine’s effects on proactive slowing.
